Generate Biomedicines

Overview

The Generate Platform combines four stages: generate, build, measure and learn. Models propose sequences, laboratories produce and characterize the proteins, and experimental results improve the models. Its applications include antibodies, enzymes and other proteins designed for specified functions.[1]

Key facts

Incorporation

The company was incorporated in Delaware in August 2018. Its jurisdiction of incorporation differs from its present headquarters in Somerville, Massachusetts.[2]

Public Launch

Flagship Pioneering publicly launched Generate. Its founding team included Avak Kahvejian, Geoffrey von Maltzahn, Molly Gibson and Gevorg Grigoryan, pursuing therapeutic candidates through relationships between protein sequence, structure and function.[3]

Michael Nally Becomes CEO

Michael Nally became CEO in 2021, overseeing expansion of the platform and clinical development. The current leadership team also includes co-founder and chief technology officer Gevorg Grigoryan and president and chief financial officer Jason Silvers.[4]

$370 Million Series B

Generate completed a $370 million Series B, its first external equity financing. Participants included Flagship Pioneering, an ADIA subsidiary, the Alaska Permanent Fund and ARCH, supporting its platform, team and preclinical programs.[5]

Amgen Research Collaboration

The companies agreed to develop protein therapeutics for five initial targets, with Amgen providing $50 million upfront. The potential agreement value was $1.9 billion plus royalties; milestone-dependent payments were not realized revenue.[6]

MD Anderson Co-Development

Generate and MD Anderson agreed to co-develop therapeutics for up to five oncology targets, including lung-cancer programs. The collaboration combines computational design, experiments and translational research, with shared R&D expenses and commercialization proceeds.[7]

CryoEM Laboratory Opened

Generate unveiled an approximately 70,000-square-foot Andover facility with four electron microscopes. CryoEM structural data help examine interactions between proteins and their targets and feed back into the machine-learning platform.[8]

Series C Announced

The September announcement reported a $273 million Series C, with new investors including Amgen and NVIDIANVIDIANVIDIA is an American semiconductor and computing technology company that provides graphics processing units, artificial intelligence infrastructure, networking interconnects, and computing software, and is listed on Nasdaq under the ticker NVDA.Open the full entry’s NVentures. Generate then described 17 preclinical and clinical programs, with proceeds supporting trial applications, clinical research and platform development.[9]

Roswell Park Cell-Therapy Collaboration

Generate and Roswell Park agreed to develop CAR T cells and armoring technologies for up to three oncology targets, including ovarian cancer. They agreed to share R&D expenses and profits from resulting commercial products.[10]

Chroma Published in Nature

Chroma research appeared in Nature, describing a model for generating proteins with specified properties. Generate released code and made weights available to academic researchers and nonprofits. Those access terms should not be described as unrestricted commercial permission.[11]

Clinical Progress and Amgen Expansion

Generate reported first-participant dosing for anti-TSLP antibody GB-0895. Amgen expanded the collaboration to a sixth program, with an undisclosed upfront payment and up to $370 million in future milestones plus royalties.[12]

Multi-Target Novartis Collaboration

Novartis and Generate agreed to develop protein therapeutics, with $65 million upfront including a $15 million equity investment. Terms also provided over $1 billion in performance-based milestones and tiered royalties. Specific targets were not disclosed.[13]

Pipeline Update

Generate reported nearly 20 programs spanning immunology, infectious disease and oncology. Alongside GB-0895, it described an anti-IL-13 candidate, a free-MMAE neutralizer and CAR T designs. Future enrollment plans in that announcement were prospective at the time.[14]

GB-0895 Phase 1 Results

Generate reported a Phase 1 study involving 96 patients with mild-to-moderate asthma, an approximately 89-day half-life and pharmacodynamic changes lasting at least six months. These findings supported further study of twice-yearly dosing, without establishing long-term clinical efficacy.[15]

SOLAIRIA Phase 3 Program Planned

Generate announced plans for SOLAIRIA-1 and SOLAIRIA-2, enrolling approximately 1,600 severe-asthma patients in comparisons of GB-0895 with placebo. The primary objective concerns exacerbations over 52 weeks; twice-yearly subcutaneous dosing remained investigational.[16]

IPO and Nasdaq Listing

Generate priced 25 million new shares at $16 each, targeting $400 million in gross proceeds before underwriting discounts and offering expenses. Underwriters also received a 30-day option to purchase additional shares.[17]

The shares began trading on Nasdaq on February 27 under the symbol GENB.[2]

Second-Quarter Update

Second-quarter collaboration revenue was approximately $6.3 million, with a $67.3 million net loss. Cash, equivalents and marketable securities totaled $457.4 million at June-end. SOLAIRIA Phase 3 recruitment was underway globally, and dosing had begun in the first GB-4362 Phase 1 cohort.[18]

Ovarian-Cancer CAR T First Dosing

GB-5267 reached first-patient dosing in a Roswell Park Phase 1 study. The autologous IL-18-armored CAR T targets MUC16 in platinum-resistant ovarian cancer. The study focuses on safety and tolerability; first dosing does not establish efficacy.[19]

Zealand Pharma Collaboration

The companies agreed to pursue multispecific proteins for obesity and metabolic disease. Generate leads research through lead-candidate selection; Zealand handles subsequent clinical development, filings and commercialization. Two initial programs carry $30 million upfront plus conditional milestones and royalties.[20]

Compact CAR Signaling Research

AI-assisted design and experimental iteration yielded a 45-amino-acid CAR intracellular domain with improved expression and sustained function in cell experiments. The preprint left in vivo performance unresolved and did not establish superiority over other design methods matched for experimental budget.[21]

The Canelé team used Claude Code to build laboratory-record and analysis tools, with scientists reviewing design rationales. Generate also announced expanded target-selection and validation work with AnthropicAnthropicAnthropic is an artificial intelligence research and product company founded in 2021 that develops the Claude models, assistant, and tools such as Claude Code, and operates as a public benefit corporation. Dario Amodei and Daniela Amodei are its co-founders, and its research covers model safety, interpretability, and steerability.Open the full entry on antigen combinations for early-stage solid tumors; the anticipated results were not yet available.[22]
